A sireline refers to the connection between an Original such as Elijah and then his/her progeny who are created using his blood. This concept applies to all the vampires and the link goes down to all the vampires which are created by each vampire that came before them. This sireline connection originated from the sophisticated spell that Esther used when she created the Originals.

So in simple terms if an Original from mostly all the vampires descended dies then the all the vampires who are part of the chain automatically die.
