It is the chain which contains the originator of a vampire line and all the other vampires who have been sired to the originators. The Originals are the first of the vampire race, they are the originators of the vampire race. If an Original is permanently killed by a White Oak Stake, his sireline would cease to exist. They would die too along with him. Esther Mikaelson who transformed her children into the vampires added a powerful link spell along with the immortality spell which would protect her children.

The sireline of Klaus was unlinked in the third season of the Originals. This is the reason why all the vampires that he sired still continue to exist even after his death in the Originals finale. Elijah was killed by the Hollow because of which his sireline died too, but he was later resurrected, but his sireline couldn’t be restored. Kol doesn’t have a sireline and that leaves Rebekah Mikaelson as the only Original with a sireline. The Upgraded and the Enhanced Original Vampires don’t have a sireline.
Another interesting fact is that Mikael, the father of the Mikaelson siblings doesn’t have a sireline. It is assumed that because of his contempt for vampires, he never sired any vampire, he only fed off them. In Season 3, Lucien stated that the vampires from all across the world were aware of the attack on Klaus. The vampires of different sirelines were at war and an attack on any of the Originals could be made.
